What is tributyltin-binding proteins (TBT-bps)?

TBT-bps is a newly and novel discovered protein that binds to tributyltin (TBT) in the blood of Japanese flounder, Paralichthys olivaceus, and is excreted from the body via the skin mucus (Shimasaki et al., 2002; Satone et al., 2008). TBT-bp1 of Japanese flounder is a glycoprotein with 42% N-glycan content and its molecular mass and isoelectric point are 46.5 kDa and 3.0, respectively. We expected that TBT accumulation in blood of flounder would attribute to TBT-bp1. The large amount of N-glycan and the amino acid sequence suggesting that TBT-bp1 may be categorized as one of lipocalin members which play an important role in transportation, detoxification and elimination of xenobiototic compounds. (Oba, et al., 2007; Satone et al., 2008). TBT-bp1 was identified in tiger puffer fish(Oba, 2010)

Response of tributyltin-binding protein by chemical exposure or stress

TBT-bp1 was up regulated by exposure of

  • TCDD in liver of medaka (Volz, 2005)
  • PCBs and PCB126 in brain of medaka (Nakayama, 2008)
  • mixture of pesticide in flounder (Marchand, 2006)
  • dimethylbenz[a]anthracene in liver of eel (Nogueiraa, 2009)
  • Cr(VI) in mummichog (Roling, 2006)
  • As in mummichogs (Gonzalez, 2010)

Also, Up regulated to fish disease

  • TBT-bps was upregulated in trubot challenged to Aeromonas salmonicida (Millan et al 2009)

Different gene expression of TBT-bp1 and TBT-bp2

  • in japanese flounder with different susceptibilities to edwardsiellosis (Yasuike, 2010)

On the other hand TBT-bp1 is down regulated by

  • exposure of Bisphenol A in fish (Lee, 2007)

Also TBT-bp2 was increased by

  • exposure of TBT-d27 (Nassef, submitted)
  • PCBs and PCB126 in brain of medaka (Nakayama, 2008).
